人生還有幾個十年?計算死亡的霍瓦特鐘


一個人的細胞都擁有相同DNA,但為什麼我們由胚胎小小的幹細胞生長成為身體不同的器官,各有不同的特徵?原因就是DNA訊息如何傳遞給幹細胞,告訴細胞要生長成為神經線、肌肉、血球,還是皮膚。

而控制哪些DNA訊息開或關就全靠DNA表層上的化學標記(甲基化Methylation)。甲基化會受外在因素如飲食、藥物甚至社交環境等而改變。科學家將DNA比喻為CD光碟,訊息永遠都記錄在光碟內,但光碟表層受損有劃痕,光碟機就不能正常讀出資料。細胞因此忘記了自己的特性,失去功能及停止分裂出新細胞,成為衰老細胞(Senescent cell)殘留在體內引發炎症或誘發腫瘤。

抗衰老科學家 Steve Horvath 於2013年發現了透過測出甲基化標記於不同年齡的變化,就能準確計算出一個人的生物年齡,甚至可以推算出他的死亡時間。人們將此命名為Horvath Clock。慶幸的是甲基化是可以改變的。這代表了衰老亦能夠被治療。科學家正研究如何重置這個生物鐘,並成功令老鼠的視神經重生,恢復視力。而透過霍瓦特時鐘,我們就可以有效量度逆齡成效。

另外,科學家發現體內的一種物質 – NMN,能夠轉化為NAD+,穩定甲基化標記,如同防止光碟上的劃痕一樣。NAD+水平會隨年齡下降,多年來實驗證實透過補充NMN,身體會產生NAD+提高細胞動能並激活長壽基因,非但沒有不良副作用,身體越變得更有活力及年輕,甚至可以預防及改善多種老年疾病。
